Magnesium Glycinate vs Citrate: A Pharmacist’s Honest Comparison

Magnesium glycinate vs citrate is one of the most common comparisons patients ask me to make in pharmacy practice. Both are popular magnesium supplements. Both are widely available. But they are not interchangeable, and picking the wrong one for your needs can mean the difference between a supplement that works beautifully and one that sends you running to the bathroom.
This comparison is my honest assessment as a pharmacist. What the evidence actually says about absorption, what each form is best for, and how to choose between them without falling for marketing claims that have no basis in science.
What These Two Forms Actually Are
Magnesium glycinate is magnesium bound to glycine, an amino acid that functions as an inhibitory neurotransmitter in the brain. Glycine has its own calming properties. It helps lower body temperature at night and signals the brain to prepare for sleep (1). The glycine molecule also serves as a carrier that helps magnesium cross the intestinal wall without drawing water into the bowel.
Magnesium citrate is magnesium bound to citric acid. Citrate comes from citrus fruits and plays a role in energy metabolism. The citrate bond makes this form highly soluble in water, which is why it dissolves easily and absorbs quickly. But that same solubility has a downside. Unabsorbed citrate pulls water into the intestine through osmosis, which is precisely how saline laxatives work (2).
This fundamental difference in chemistry drives everything else. Glycinate is built for calm. Citrate is built for movement.
The Absorption Question: What the Evidence Actually Shows
If you have spent any time reading about magnesium, you have probably seen claims that one form is dramatically more absorbable than another. The truth is more nuanced and more interesting.
A 2021 systematic review that compared the bioavailability of different magnesium preparations found that organic forms of magnesium, which includes both glycinate and citrate, are more bioavailable than inorganic forms like magnesium oxide (3). The review also noted that the percentage of magnesium absorbed from citrate is dose dependent. Higher doses mean a lower fraction absorbed.
A 2019 study published in Biological Trace Element Research investigated how different magnesium compounds distribute into tissues (4). This study examined both amino acid bound compounds like magnesium glycinate and organic acid bound compounds like magnesium citrate. The researchers found that magnesium citrate increased muscle and brain magnesium levels in a dose independent manner, meaning it raised tissue levels reliably even at different doses.
Here is the honest truth that most marketing material will not tell you. There has never been a published clinical trial that directly compares magnesium glycinate and magnesium citrate head to head in humans for absorption (5). No such study exists. When manufacturers claim that glycinate is several times more absorbable than citrate, they are making statements that have no comparative human data behind them.
What the existing research does suggest is that both are well absorbed organic forms. Citrate may actually have a slight edge in some absorption metrics, but the difference between the two is likely small and clinically less important than the difference between either of them and magnesium oxide (3). Side Effects: Where the Real Difference Lives
In clinical practice, absorption is not the deciding factor between these two forms. Tolerability is.
Magnesium citrate is classified by the FDA as a saline laxative. Its official drug label states that it is indicated for the relief of occasional constipation and generally produces a bowel movement within 30 minutes to 6 hours (2). This is not a side effect in the conventional sense. It is the primary mechanism of action for citrate. The unabsorbed portion of the dose pulls water into the colon, which stimulates bowel movement. For someone who is constipated, this is a benefit. For someone who is taking magnesium for sleep or anxiety and has no issues with constipation, this is a problem.
Magnesium glycinate does not have this laxative effect in most people. Because it is bound to glycine rather than an organic acid, it does not create the same osmotic pull in the intestine. Glycinate is generally considered the gentlest form of magnesium on the stomach and the least likely to cause digestive side effects (6). For people who have tried magnesium before and experienced diarrhea, switching to glycinate often solves the problem.
The practical clinical difference is this. If you need magnesium for sleep, anxiety, or general supplementation and you have a sensitive stomach, glycinate is almost always the better choice. If you are dealing with constipation and also want magnesium repletion, citrate can serve both purposes at once.
Which Form Is Best for Sleep and Anxiety
Magnesium glycinate has developed a strong reputation as the sleep form of magnesium. There is good mechanistic reasoning behind this. The glycine component of glycinate has its own calming effects. Glycine acts as an inhibitory neurotransmitter in the brainstem and spinal cord, and oral glycine supplementation has been shown to improve subjective sleep quality (1). Magnesium itself supports GABA receptor function, as I have covered in my GABA article, and helps regulate the body’s stress response system.
A 2025 randomized, placebo controlled trial examined magnesium bisglycinate supplementation in adults reporting poor sleep (7). The study found that magnesium bisglycinate modestly improved insomnia severity compared to placebo, though the effect was small. Most of the benefit was seen in people whose baseline dietary magnesium intake was low.
For anxiety, the evidence is suggestive but not definitive. A 2017 systematic review of 18 studies found that magnesium supplementation was associated with reductions in subjective anxiety, particularly in people who were vulnerable to stress (8). However, most of these studies did not use magnesium glycinate specifically. The anxiety benefits of magnesium appear to be linked to correcting deficiency rather than a drug-like anxiolytic effect in people with normal magnesium levels.
Magnesium citrate has far less direct evidence for sleep or anxiety. Its main role is digestive, and the laxative effect can actually disrupt sleep if it causes nighttime bowel urgency. I would not recommend citrate for someone whose primary goal is better sleep unless they also need help with constipation.
Which Form Is Best for Constipation
Magnesium citrate is one of the most widely used over the counter saline laxatives in the United States. Its effectiveness for relieving occasional constipation is well established, and it works quickly. The standard dose for constipation is 6.5 to 10 fluid ounces of the liquid formulation, which provides roughly 1.7 grams of magnesium citrate per fluid ounce, generally producing a bowel movement within 30 minutes to 6 hours (2).
Magnesium glycinate is not an effective laxative at standard doses. The glycine chelate does not produce the osmotic effect that citrate does, and taking high enough doses of glycinate to trigger a bowel movement would likely cause other forms of GI discomfort. If constipation is your primary concern, citrate is clearly the better choice. If you are considering using magnesium for both constipation and sleep, taking citrate in the morning rather than at night can help you get the digestive benefit without risking sleep disruption.
Other Health Benefits Worth Knowing
Both forms of magnesium support bone health. Magnesium is essential for bone mineral density, and supplementation has been shown to suppress bone turnover in postmenopausal women (9). The benefit appears to be related to magnesium status rather than the specific form used.
Both forms also play a role in blood sugar regulation. Magnesium supplementation has been shown to modestly lower fasting blood sugar and improve insulin sensitivity in people with or at risk for type 2 diabetes (10). Again, the benefit is linked to magnesium itself, not the specific salt form.
Magnesium citrate has been studied for migraine prevention, with some evidence suggesting benefit particularly for people with low magnesium levels (11). The proposed mechanism involves magnesium’s effects on blood vessel tone and neurotransmitter release. Both forms should provide this benefit if magnesium status is low.
The Honest Cost Comparison
Magnesium citrate is generally less expensive per dose than magnesium glycinate. This is partly because citrate has been on the market longer and is produced at larger scale, and partly because glycinate requires a more complex manufacturing process to chelate magnesium to glycine.
The price difference is not enormous for most brands, but over months or years of daily use, it adds up. If cost is a significant factor and you tolerate citrate well, it is the more budget friendly option for maintaining magnesium status. If you are sensitive to citrate’s laxative effects, the extra cost of glycinate is worth it for the tolerability.
How to Choose Between Them
The decision comes down to three questions.
First, what is your primary goal? For sleep, anxiety, or stress, glycinate is the better choice. It provides the additional calming benefit of glycine and is far less likely to cause digestive disruption at sleep doses. For constipation or occasional digestive sluggishness, citrate is the clear winner.
Second, how sensitive is your stomach? If you have tried magnesium before and experienced loose stools, switch to glycinate. It is the gentlest form available and tolerability in real world use is excellent.
Third, what is your budget? Citrate is less expensive and widely available. If you tolerate it well and your main goal is simply to maintain adequate magnesium intake, citrate is a perfectly reasonable choice.
A practical approach many patients take is to keep both forms on hand. Glycinate in the evening for sleep and relaxation. Citrate as needed for occasional constipation. They serve different purposes, and there is no harm in using both as long as you track your total magnesium intake.
My Pharmacist’s Verdict
I am asked about magnesium glycinate vs citrate almost daily, and my answer almost always starts with the same question: what are you trying to achieve?
For sleep, anxiety, and sensitive stomachs, magnesium glycinate is the better choice. The glycine component adds a small but real calming benefit, and the tolerability advantage is significant. Most patients who switch from citrate to glycinate for sleep report fewer digestive side effects.
For constipation, magnesium citrate is the appropriate choice. It is an FDA recognized saline laxative with decades of clinical use behind it, and it also provides magnesium repletion at the same time.
Both forms are well absorbed organic magnesium compounds. The marketing claims that glycinate is dramatically more bioavailable are not supported by head to head comparative data in humans. The real difference between these two forms is not absorption. It is tolerability and clinical application.
If you are new to magnesium supplementation and are unsure where to start, begin with glycinate at 200 to 300 mg of elemental magnesium in the evening. It is the gentlest introduction and the most versatile for common goals like sleep and stress. If you tolerate it well and want to experiment with citrate later, you can always switch.
If you take prescription medications, speak with your pharmacist before starting magnesium. Magnesium can interact with certain antibiotics, bisphosphonates used for osteoporosis, and some blood pressure medications. The interaction is manageable with proper spacing, but it needs to be addressed proactively.
